ABSTRACT

Aim: The objective of the study was to assess the effect of dietary incorporation of different feed attractants viz. tubifex, earthworm and fish meal on growth and survival of O. bimaculatus (pabda fish) fry. Methodology: Fifty numbers of fifteen days weaned fry (0.105±0.008g; 2.14±0.14cm) were stocked into each aerated aquariums (30.0 x 15.0 x 15.0 cm) following a completely randomized design (CRD) consisting of four treatments including the control with three replicates each. Four iso-nitrogenous purified diets were prepared including three treatment diets with attractants (5%) by replacing an appropriate amount of casein and cellulose and fed to the fishes twice daily. Results: Results from the study showed that growth of fry as measured by final weight, weight gain, mean daily weight gain and specific growth rate of fries were significantly (p<0.05) higher in tubifex supplemented group and lower in control group. Survival was significantly higher in tubifex supplemented group (42.66±1.3%) and lower in control group. Moreover, other yield parameters such as total biomass, condition factor and performance index also followed the same trend as survival. Further, higher protease activity was found in control group (2.233±0.038 units mg protein-1 min-1) followed by earthworm (0.354±0.031 mg protein-1 min-1) supplemented group (p<0.05). The lipase and amylase activity were higher (p<0.05) in tubifex and control groups, respectively. Interpretation: It could be concluded that the dietary supplementation of tubifex at 5% inclusion level could be a promising aquaculture feeding strategy for pabda fish due to its rich nutrient content, increased feeding stimulation and more palatability.

ABSTRACT

To explore the effect and mechanism of Astragali Radix on growth and immunity of Whitmania pigra, 0, 0.01%, 0.03%, 0.05%, 0.07%, 0.09% of Astragali Radix were added to the daily feeding of Wh. pigra. After 60 days of feeding, the growth performance, activities of digestive enzyme and anti-reverse enzyme, inner quality, the expression levels of GH, IGF-1 and digestive enzyme-related genes were measured. Meanwhile, the effects of heat stress on the living conditions of Wh. pigra were observed and counted, and the expression levels of HSP70 and immune related genes were measured. The results showed that the final weight, weight gain rate, specific growth rate, activities of digestive enzyme and anti-reverse enzyme, the expression levels of GH, IGF-1 and digestive enzyme-related genes in the Astragali Radix group were higher than those in the control group, and with the increase of Astragali Radix concentration, the above-mentioned indexes increased initially and then decreased, and significantly higher in the 0.05% of Astragali Radix group than in the other groups (<0.05). There was no significant difference in the inner quality of Wh. pigra between the Astragali Radix and control groups. The survival rate of Wh. pigra was negatively correlated with heat stress treatment duration. With the prolongation of heat stress treatment duration, the expression levels of HSP70 and immune related genes were increased first and then decreased, and peaked at 24 h. The survival rate and the expression levels of HSP70 and immune related genes in the Astragali Radix group were higher than those in the control group, and was significantly higher in the 0.05% of Astragali Radix group than in the other groups (<0.05). In conclusion, Astragali Radix can increase the activities of digestive enzyme and anti-reverse enzyme, the expression levels of related genes, growth performance, and immunity to heat stress of Wh. pigra. It is suggested to add 0.05% of Astragali Radix in the actual production of Wh. pigra to improve the production profit.

ABSTRACT

Objective To investigate the effect of Shenling-Baizhu powder on gastrointestinal hormone,digestive enzyme and inflammatory factors in D-IBS rats.Methods A total of 50 Wistar rats were randomly divided into the normal group,the model group,the Shenling-Baizhu powder large dose group,medium dose group and small dose group,10 rats in each group.D-IBS rat model was induced by high lactose diet and restraint stress except normal group.At the same time,the Shenling-Baizhu powder groups received Shenling-Baizhu powder (large dose group:9 g/kg,medium dose group:3 g/kg,small dose group:1 g/kg),normal group and model group received equal volume of normal saline.Malate dehydrogenase (MDH),lactate dehydrogenase (LDH) and disaccharidase activities were detected from small intestine of rats in each group.Plasma vasoactive intestinal peptide (VIP) and substance P (SP) activities were measured by radioimmunoassay.TNF-α and IL-10 levels in serum were determined by ELISA.Results Compared with the model group,the model group,the ratio of serum TNF-α/IL-10 (0.31 ± 0.15,0.34 ± 0.21,0.41 ± 0.15 vs.0.74 ± 0.19) significantly decreased in large,medium and low dose groups (P<0.05).The disaccharidase activities in the small intestinal mucosa (38.44 ± 11.30 U/mg,44.77 ± 16.46 U/mg,42.58 ± 13.02 U/mg vs.23.23 ± 6.85 U/mg) significantly increased in the large,medium and low dose groups (P<0.05).LDH activities in the small intestinal mucosa (1 863.61 ± 1 019.28 U/mg,1 136.13 ± 631.89 U/mg vs.416.50 ± 223.94U/mg) significantly increased in large,medium dose groups (P<0.05).Plasma SP level (22.88 ± 10.17 pg/mg vs.31.86 ± 7.81 pg/mg) significantly decreased in large dose group (P<0.05).Conclusions Shenling-Baizhu powder may increase the activities of LDH,MDH and disaccharidase,inhibit the abnormal secretion of SP and regulate of Th1/Th2 immune imbalance to relieve diarrhea.

ABSTRACT

OBJECTIVE:To observe the efficacy and safety of proton pump inhibitors combined with Compound digestive en-zyme tablet in the treatment of epigastric pain syndrome with abdominal distension. METHODS:156 patients with epigastric pain syndrome with abdominal distension were randomly divided into control group(75 cases)and observation group(81 cases). Con-trol group was given 40 mg Esomeprazole enteric-coated tablet for once half an hour before breakfast;observation group was addi-tionally given 243.6 mg Compound digestive enzyme tablet immediately after meal,3 times a day. The treatment course for both groups was 2 weeks. Clinical efficacy,clinical symptom score,pepsinogenⅠ(PGⅠ),pepsinogenⅡ(PGⅡ),2,4 h postprandial gastric emptying rate before and after treatment,and incidence of adverse reactions in 2 groups were observed. RESULTS:The to-tal effective rate in observation group was significantly higher than control group,the difference was statistically significant (P0.05). Before treatment, there were no significant differences in the clinical symptom score,PGⅠ,PGⅡand 2,4 h postprandial gastric emptying rate(P>0.05). After treatment,clinical symptom score in 2 groups were significantly lower than before,and observation group was lower than control group;PGⅠ and PGⅡ in observation group were significantly higher than before and control group,the differences were statistically significant(P0.05). CONCLUSIONS:The efficacy of pro-ton pump inhibitors combined with Compound digestive enzyme tablet is superior to the proton pump inhibitors in the treatment of epigastric pain syndrome with abdominal distension,with similar safety.

ABSTRACT

The biological characteristics, oxygen consumption, oxygen consumption rate, and activities of amylase, lipase and protease of Whitmania pigra at different temperature were studied by using direct observational method, the still water method and 3, 5-dinitrosalicylic acid colorimetry, right-nitrophenyl palmitate ester(ρ-NPP)colorimetry and folin-phenol method.The results revealed that with decreasing water temperature, the daily activity and the daily feeding ration were decreased. As the temperature was lowered to 4 ℃, the head and tail of Wh.pigra curved, showing a crescent-shape without feeding and daily activity. Oxygen consumption, oxygen consumption rate and digestive enzyme activities reduced along with temperature drops. The downward trend slowed below 10 ℃, began to stabilize below 4 ℃ and doesn't change with the decrease of temperature since then. During the 40 days treatment at 4 ℃, the changes of amylase were not significant, the lipase and protease activity decreased at the 20th day, and the lipase showed an slightly increase after the decrease and finally remained at a low level.In conclusions, the pivotal temperature of hibernation of Wh.pigra is 4 ℃ and the crescent shape can be considered as a symbol of hibernation.

ABSTRACT

The effects of stocking density and exchanging water frequency on growth, digestive enzyme activity, anti-oxidative enzyme and inner quality of Whitmania pigra Whitman were evaluated with corresponding measures. The results showed that the eventual biomass, specific growth rate, gained weight rate, activities of amylase, lipase, protease, SOD, CAT, and ALP correlated positively with stocking density and negatively with exchanging water frequency (P<0.05). Exchanging water frequency had negative correlation with ammonia nitrogen, nitrite, and hydrogen sulfide while revealed positive correlation with dissolved oxygen in the water. Stocking density and exchanging water frequency showed no significant effects on the contents of moisture, total ash, and acid-insoluble ash. It suggested that the optimum stocking density was 7.5 million per hectare and the appropriate exchanging water interval was 72 h.

ABSTRACT

OBJECTIVE:To observe therapeutic efficacy and safety of Compound digestive enzyme capsule in the treatment of dyspepsia. METHODS:154 patients with dyspepsia were selected and randomly divided into observation group and control group, with 77 patients in each group. Control group received routine treatment,Polyzyme tablet 600 mg,tid,30 min after meal;observa-tion group was additionally given Compound digestive enzyme capsule 600 mg,tid,30 min after meal. Treatment course of 2 groups lasted for 2 weeks. The improvement of dyspepsia,clinical efficacy,clinical manifestation score,improvement duration and ADR were observed in 2 groups after treatment. RESULTS:The total effective rate of observation group(89.61%)was significant-ly higher than that of control group(74.03%),with statistical significance(P0.05). CONCLUSIONS:Compound digestive enzymes capsule has good effect on dyspepsia with less ADR.

ABSTRACT

Objective Primary myocardial cell culture , as an important technique for in vitro experiment , provides an essen-tial basis for the establishment of models of cardiovascular diseases .This study aimed to investigate the influence of digestion time on primary myocardial cell culture and to find some simple primary culture methods for obtaining myocardial cells with high survival rate , purity, and activity. Methods We used only one collagenase for repeated digestion of myocardial tissue for 8 and 10 minutes, re-spectively , and obtained high-purity cardiomyocytes by differential adhesion and Brdu inhibition .We observed the morphological chan-ges of the cardiomyocytes under the optical microscope , measured their vitality according to the beating frequency in different regions , and counted the living cells after typan blue staining. Finally, we established a model of myocyte hypertrophy for verification of cell activity and compared the cell surface area between the experimental ( angiotensinⅡ) and control ( 10%FBS DMEM [H +] medium ) groups. Results After cultured for 48 hours, the primary cardimyocytes in the 10 min group mostly extended pseudopodia , in-terwoven into a network and gradually forming cell clusters , with obviously powerful beating and contraction .After isolation and purifi-cation, the average survival rate of the cells was >90%, significantly higher in the 10 min group than in the 8 min ([95.4 ±0.8]%vs [93.0 ±0.8]%, P95%in both of the groups.After cultured with angiotensinⅡfor 48 hours, the 10 min group showed a significantly increased level of atrial natriuretic peptide (ANP) (20.8 ±0.67 vs 15.8 ±0.48, P<0.05) and cell surface area ([34.77 ±8.43] μm2 vs [14.11 ±4.29] μm2, P<0.05) as compared with the 8 min group. Conclusion Primary culture of cardimyocytes with 8 min digestion is simple and time-saving, and what is most important , can obtain high-quality cardiomyocytes .

ABSTRACT

To evaluate the effects of supplemental probiotics and xylo-oligosaccharide on performance,digestive enzyme activities,blood index and intestinal microflora.Two hundred and forty crossbred early weaned PIC piglets with an average initial weight of (6.83 + 0.9) kg,weaned at (21 ± 2) d of age,were divided into four groups.Control Ⅰ (positive diet,with no probiotics and xylo-oligosaccharide but more fish meal,whey powder and little soybean meal),Control Ⅱ (negative diet,with no probiotics and xylo-oligosaccharide but less fish meal,whey powder and more soybean meal),Negative diet + 0.035 % probiotics,negative diet + 0.002% xylo-oligosaccharide.The results of theses studies suggested that whey powder and fish meal from 5.50% to 2.50%,soybean meal from 22.50% to 26.50% could result in decreasing of alimentary canal enzyme activity,blood sugar,total serum protein and the quantities of Bacillus in appendix(P<0.05),and raising of serum urea nitrogen,the quantities of Escherichia coli in appendix,diarrhea indexes and Feed/Gain(P<0.05).Addition of probiotics and xylo-oligosaccharide could significantly improve the activities of alimentary canal protein,lipase and amylase(P<0.05),decrease Feed/Gain and the levels of blood sugar and decrease the levels of serum urea nitrogen and improve protein equilibrium in blood and energy metabolism.The quantities of bacillus were improved,Escherichia coli were decreased in appendix(P<0.05).Diarrhea indexes were decreased(P<0.05).Whether adding probiotics and xylo-oligosaecharide or not could not affect ADG and ADFI (P>0.05).Addition of probiotics and xylo-oligosaccharide could improve digestive function,beneficial microbial population of post-alimentary canal,Feed/Gain and prevent the diarrhea of piglets.

ABSTRACT

Objective To study the influence of probiotics on the gut microflora,digestive enzymes and small intestinal propulsive rate of rats with severe head injury. Methods The rat model of severe head injury was prepared, SD rats were randomly divided into group A (enteml nutrition) , group B (enteral nutrition plus probiotics) , and group C (normal diet) . The intestine mucesa and faeces were collected on the third day,7th day and 14th day after head injury in order to detect gut microflora,digestive enzymes and small intestinal propulsive rate. Results Compared with group C, the number of Lactobacillus and Bacillus bifidus group in group A and group B declined in two weeks after trauma, however, the number of Escherichia coli increased significantly. Bacillus bifidus amounts of group A were significantly lower than that of group B at every time point, the diversity of Escherichia coli was opposite. No difference was seen in content of Lactobacillus 7 days after trauma in group A and B. The level of disaecharidases, Na+- K+- ATPase and small intestinal propulsive rate declined significantly at every time point, compared with group C.Though the contents of digestive enzymes was higher in group B than that of group A, and small intestinal propulsive rate was higher in group A and B than that in group C, but the two groups showed no difference. Conclusions Probiotics can alleviate the in-testine microflora disorder,modulate the activities of digestive enzymes, therefore lessen malabsorption of rats with severe head injury.

ABSTRACT

Chestnut as a Food Allergen: Identification of Major Allergens To evaluate the clinical significance of chestnut as a food allergen in Korea, skin prick test and ELISA were done in 1,738 patients with respiratory allergies. To identify the IgE binding components, IgE-immunoblotting, 2D IgE-immunoblotting and MALDITOF were performed. To observe the effects of digestive enzymes and a boiling treatment, simulated gastric fluid (SGF) and simulated intestinal fluids (SIF) were incubated with chestnut extracts, and IgE-immunoblotting were then repeated. Skin prick test revealed that 56 (3.2%) patients showed more than 2+ of allergen to histamine ratio to chestnut. Among the 21 IgE binding components, 9 bands were found in more than 50% of the sera tested and the 24 kDa protein had the highest binding intensity. The amino acid sequence of the 24 kDa protein (pI 6.3) had homology with legume protein of oak tree. SGF, SIF and boiling treatment were able to suppress the IgE binding components. In conclusion, chestnut ingestion was shown to induce IgE mediated responses with a 3.2% sensitization rate. Twenty one IgE binding components and one new allergen (the 24 kDa protein) were identified. Digestive enzymes and boiling treatment were able to decrease the allergenic potency.
